Byrsonima sericea leaves are extensively used in folk medicine in Brazil against gastric disorders. This study investigated the chemical constituents of B. sericea leaf ethanolic extract (BSLE) and its potential gastroprotective activity, with its possible mechanism of the action using ethanol to induce gastric mucosal damage in mice. The phytochemical analysis was carried out to identify the active constituents present in the extract, and the HPLC analysis was performed for the identification of flavonoids. BSLE at oral doses of 125, 250 and 500 mg/kg markedly attenuated the ethanol-evoked gastric lesions by 53.2, 84.9 and 87.6 %, respectively. The BSLE (250 mg/kg) prevented the depletion of gastric mucus and gastric mucosal nonproteic-sulfhydryl groups,... |

Extracts of Echinodorus grandiflorus obtained from dried leaves by three different techniques were evaluated by bacterial lysogenic induction assay (Inductest) in relation to their genotoxic properties. Before being added to test cultures, extracts were sterilized either by steam sterilization or ultraviolet light. Only the extracts prepared by infusion and steam sterilized have shown genotoxic activity. The phytochemical analysis revealed the presence of the flavonoids isovitexin, isoorientin, swertisin and swertiajaponin, isolated from a genotoxic fraction. They were assayed separately and tested negative in the Inductest protocol. The development of browning color and sweet smell in extracts submitted to heat, prompted further chemical analysis in... |

Production of baicalein, baicalin and wogonin by liquid culture of Scutellaria baicalensis cells derived from the plant root was studied. The maximum production obtained were 119 mg/L of baicalein at two week, 1372 mg/L of baicalin at eight week, and 14 mg/L of wogonin at two week. In addition, the production of baicalin was drastically increased to 1000 mg/L level at 3-week culture, and the extremely high production rate (339 mg/L•week) was obtained. In the comparison of total antioxidative activities among baicalein, baicalin and wogonin, evaluated by thiocyanate method, it was suggested that the location of hydroxyl groups both at 5- and 6-position contributed to enhancement of radical scavenging activity, and/or methoxylation at 8-position diminished... |

Blast, caused by Pyricularia oryzae, has become a significant disease threat to wheat (Triticum aestivum L.) in Brazil. This study aimed to investigate at the histochemical level if silicon (Si) could enhance the production of flavonoids in the leaves of wheat plants in response to P. oryzae infection. Plants from the Aliança cultivar, which are susceptible to blast, were grown in hydroponic cultures containing 0 (-Si) or 2 mM of Si (+Si) and inoculated by spraying a conidial suspension of P. oryzae (1 × 105 conidia mL−1) on all adaxial leaf surfaces of plants at 60 days after emergence (growth stage 65). The fourth and fifth leaves of each plant were used to evaluate blast severity at 24, 36, 48, 72 and 96 h after inoculation (hai). At 96 hai, leaves were... |

ABSTRACT Thioredoxin reductases (TrxRs) are selenocysteine-containing flavoenzymes that reduce Trxin NADPH-dependent manner. In the view of the direct vital role of TrxR in a wide range of biochemical and physiological processes, methods to inhibit this enzyme are clinically important. TrxR has recently emerged as a new candidate in anticancer drug investigations because of overexpression in tumorous cells. In this study, TrxR from chick liver was purified 94.6-fold with a yield of 4.86% and a specific activity of 0.19 EU/mg. KM and Vmax values of TrxR for DTNB were calculated as 0.9 mM and 0,03 EU/mL, respectively. Then, the effects of the flavonoids hesperidin, naringenin, chlorogenic acid, ferulic acid, naringin, 3,4-dihydoxybenzoic acid, and ellagic... |

The high morbidity, high socioeconomic costs and lack of specific treatments are key factors that define the relevance of brain pathology for human health and the importance of research on neuronal protective agents. Epidemiological studies have shown beneficial effects of flavonoids on arteriosclerosis-related pathology in general and neurodegeneration in particular. Flavonoids can protect the brain by their ability to modulate intracellular signals promoting cellular survival. Quercetin and structurally related flavonoids (myricetin, fisetin, luteolin) showed a marked cytoprotective capacity in in vitro experimental conditions in models of predominantly apoptotic death such as that induced by medium concentrations (200 µM) of H2O2 added to PC12 cells in... |

ABSTRACT Leishmaniasis and Chagas disease affect millions of people in tropical and subtropical regions. Drugs used currently to treat such diseases often present undesirable side effects and low efficiency. The aim of this work was to identify extracts and isolated compounds from the genus Lippia with leishmanicidal and trypanocidal activity. Fifteen extracts from different plant parts of Lippia species with partially known chemical compositions, four partition fractions, six compounds and a mixture of four interconverting flavanones previously isolated from Lippia salviaefolia and Lippia lupulina were assayed in vitro towards epimastigote forms of Trypanosoma cruzi and promastigote forms of Leishmania amazonensis. The root extract of L. lupulina had... |

Abstract: The objective of this work was to evaluate the performance, body composition, and physiological state of tambaqui (Colossoma macropomum) fingerlings fed diets containing different levels of green tea (Camellia sinensis) extract (GTE). A completely randomized design was used with five treatments, different levels of GTE (0.00, 0.05, 0.30, 0.55, and 0.80 g kg-1) and three replicates. The diets were offered to fingerlings twice a day, for 90 days. The increase of dietary GTE levels promoted a positive quadratic effect on body lipids, hepatic lipids, retention of body lipids, glucose, and total triglycerides, and the minimum values of these variables were estimated with 0.61, 0.54, 0.73, 0.60, and 0.30g kg-1 GTE in the diet, respectively. There was a... |

The objective was to evaluate the effect of propolis-based products (PBP) on performance, digestibility, microbial production and carcass characteristics of feedlot young bulls. Twenty-seven crossbred young bulls were used, with 353 ± 28 kg of body weight in a completely randomized experimental design, divided in three treatments: two diets with PBP with different dosages (PBP1= 0.018 mg g-1 and PBP2= 0.036 mg g-1 of total flavonoids in chrysin) and control diet (CON). To determine total digestibility, the indigestible dry matter was used as an internal marker, while microbial production was estimated from purine derivatives in urine, collected by the spot method. The evaluated carcass characteristics were: hot carcass weight, dressing percentage,... |

Resistance to chemotherapy in cancer cells is mainly mediated by overexpression of P-glycoprotein (Pgp), a plasma membrane ATP-binding cassette (ABC) transporter which extrudes cytotoxic drugs at the expense of ATP hydrolysis. Pgp consists of two homologous halves each containing a transmembrane domain and a cytosolic nucleotide-binding domain (NBD) which contains two consensus Walker motifs, A and B, involved in ATP binding and hydrolysis. The protein also contains an S signature characteristic of ABC transporters. The molecular mechanism of Pgp-mediated drug transport is not known. Since the transporter has an extraordinarily broad substrate specificity, its cellular function has been described as a "hydrophobic vacuum cleaner". The limited knowledge... |
